This project is aimed to test candidate genes and genetic pathways which may supress the toxic effects of mutant huntingtin in the brain. Our laboratory is applying high-speed BAC transgenesis to generate mice that express candidate suppressor genes in the brain. We will breed these HD suppressor mice with the BACHD mice to test their efficacy in preventing mutant huntingtin toxicity in vivo. Identification of effective genetic suppressors may provide novel mechanistic insights and therapeutic targets for HD.
